capsules (protonic pump inhibitor) antiacidic, antiulcerous
1 capsule contains 20 mg of omeprazol.
Omeprazol is a
protonic pump inhibitor that reduces the gastric acidity by blocking the
enzymatic system hydrogen-kalium adenosine triphosphate (“protonic
pump”) of the gastric parietal cells. In combination with antibiotics,
this drug is irreplaceable in eradication of Helicobacter pylori.
Omeprazol is especially effective in cases resistant to other
antisecretory drugs. In therapeutic doses, omeprazol maintains the pH of
the stomach around 3, in duration of 16 to 18 hours.
ventricular ulcer, “resistant ulcer”, oesophageal reflux,
Zolinger-Elison syndrome, eradication of Helicobacter pylori in
combination with other antimicrobic substances.
There are no known contraindications to omeprazol.
Dosage and administration
Omeprazol should be
administered prior meal, preferably prior having a breakfast. Capsules
have to be swollen with a little water.
Preferably, 1 capsule
of 20 mg, once a day, in 2 weeks. In cases of an ulcer resistant to
other drugs, dose might increase to 40 mg/day.
Preferably, 1 capsule
of 20 mg, once a day, in 8 weeks. In severe cases, dose might increase
to 40 mg, once a day.
Preferably, 1 – 2 capsules (20 mg – 40 mg/day), 4 to 8 weeks.
Preferably, 2 capsules
of omeprazol (40 mg), once a day, in 4 weeks. If improvement is not
achieved, therapy continues for 4 more weeks.
Initial dose is 60 mg,
once a day (3 capsules of omeprazol). Dose should adjust to individual
needs, treatment continues as long as clinically necessary. Therapy
might administer for more than 5 consecutive years. If the daily dose of
omeprazol is over 80 mg, 2 divided doses are to be administered.
Highest daily dose is not allowed to be over 120 mg/day.
Eradication of Helicobacter pylori
In treatment of
infections with H.pylori, besides use of Omeprazol, it is necessary to
administer some other antibiotic too, respectively triple therapy:
- 2 times a day 20 mg of Omeprazol,
- 2 times a day 1000 mg of amoxicillin,
- 2 times a day 500 mg metronidazol.
This combination is
administered for 7 days, whilst Omeprazol continues for 14 more days.
Omeprazol is well
tolerated by patients. Rare side effects that can occur are as follows:
diarrhoea, constipation, abdominal pain, headache, dizziness, rash,
urticaria and pruritus. In rare cases, are observed hematologic
disorders such follows: leukopenia, neutropenia, thrombocytopenia,
interstitial nephritis. Changes in hepatic enzymes, gynecomastia,
impotence, and stomatitis.
